These are the services provided by Translational Proteomics Center (TPC):

  • Depletion of most abundant proteins
  • BCA (Total Protein Quantitation)
  • Immunoprecipitation
    • >> Investigators must provide the antibodies and the lysis buffer.
  • Sample Preparation:
    • Acetone Precipitation
    • SDS PAGE
    • Coomassie
  • Sample Processing for Mass Spectrometry:
    • Reduction and Alkylation
    • Enzymatic digestion in-gel
    • Enzymatic digestion in solutionPeptide Extractions
    • Peptide cleaning (when required)
  • Protein Identification:
    • Analysis by LC-MS/MS 
    • Proteome Discoverer 2.5 Data Search
  • Quantitative Proteomics:
    • Peptide tagging (TMT labeling)
    • Analysis by LC-MS/MS (Easy nLC 1200/Q-Exactive Plus)
    • Peptide Fractionation
    • Proteome Discoverer 2.1 Data Search

 

Notes:

  • An initial consultation with Dr. Loyda M. Meléndez and Staff is required to determine the experimental approach. 
  • Quantitative Proteomics by TMT Labelling requires a supply kit included in the final Invoice.
  • Requires Bio-informatics analysis. Data generated by Mass Spectrometry will be transferred directly to Dr. Abiel Roche’s facility. Additional costs apply that should be coordinated with Dr. Roche.  Analysis cost will be included in the Proteomics Invoice.
  • All presentations, publications, grants submissions or any other outcome achieved containing the TPC facility acknowledgement must be notified to the staff. These are reported to NIH every trimester and is a key component for maintenance of TPC.
  • All protocols performed in the TPC facility reported in any scientific forum must be revised by a TPC staff to ensure accuracy of the information presented containing the TPC facility acknowledgement.
  • When a TPC director and staff actively participates the experimental design, protocol development, manuscript writing and revision, authorship will be granted. Manuscripts and Publications are reported to NIH every trimester and is a key component for maintenance of TPC.
